During the last blues class we practiced 12-bar blues riffs using eighth notes, triple-eights, quarter notes, half notes, and whole notes. Its best to mix them up for variety, but concentrate on the target notes (the 1-4-5 root notes). You can even hang in the 1 root note while the blues progresses through the 4 and the 5 and it still sounds good.
