To play blues, you improvise, and play different riffs and play scale patterns, so you when looking at a song like this, you want to play the 1 5 4 progression, and also learn the scale patterns that fit that.
Everybody went around the room playing the lead. The lead is really the melody line that a singer would sing. You can take your lead by (1) attacking the target notes and (2) building off the target notes, and using combinations of riffs and the scale patterns.
Moral of the story: You can’t approach a song like this the same way your approach a bluegrass song. You have to look at your pentatonic scales, have to understand your target notes as 1 4 5 progressions, and you can then also learn the lead as the melody line. If you know your scales you can play any blues song by developing riffs.
Find your target notes, and your scales, because that is where you can build your riffs.
