Remember that the 5th string is the short one at the top right next to your thumb, unless you play you banjer upside down like the guy I saw in Tennessee this summer. He was left handed but played a righthanded banjer. Played fiddle and guitar lefty also.

Did you mean fretting the fifth string with your thumb, or picking the fifth with your thumb? I think the answer is yes to both.
